cUDG enzyme catalyzes the hydrolysis of uracil base containing uracil DNA chain and n-glycoside bond of sugar phosphate skeleton, releasing free uracil. This enzyme can act on single - or double-stranded DNA containing dU and is inactive for RNA. The principle of pollution prevention is: in the PCR reaction, dUTP is substituted for dTTP and incorporated into DNA to form the PCR amplification product containing dU bases. cUDG enzyme can selectively break the glycoside bond of dU bases in single and double stranded DNA and degrade the PCR amplification product containing dU.

Activity |
With uracil in DNA as the substrate, the enzyme amount required to release 1nmol uracil free within 1 hour at 37°C in 70mM tris-HCl, pH7. 5 (at 25°C), 10mM NaCl, 1mM EDTA, and 100ug/ml BSA reaction solution was defined as 1 active unit (U).
