Leading Biology Launched Human FGL1 Protein


Richmond, CA, Apr 1st, 2019 - Leading Biology announced immediate availability of human FGL1 protein.


Fibrinogen-like protein 1 (FGL1) is a member of the fibrinogen family. FGL-1 is homologous to the carboxy terminus of the fibrinogen beta- and gamma- subunits which contains the four conserved cysteines of that are common to all members of the fibrinogen family. However, FGL-1 lacks the platelet-binding site, cross-linking region, and thrombin-sensitive site which allow the other members of the fibrinogen family to aid in fibrin clot formation. This protein may play a role in the development of hepatocellular carcinomas. Four alternatively spliced transcript variants encoding the same protein exist for this gene.


Recently a study proved the relationship between FGL1 and LAG-3. They demonstrated that fibrinogen-like protein 1 (FGL1) is a major LAG-3 functional ligand independent from MHC-II. FGL1 inhibits antigen-specific T cell activation, and ablation of FGL1 in mice promotes T cell immunity. Blockade of the FGL1-LAG-3 interaction stimulates tumor immunity and is therapeutic against established mouse tumors in a receptor-ligand inter-dependent manner. Their findings revealed a new immune evasion mechanism and had implications for the design of cancer immunotherapy[1].

Interleukin 6 (IL-6) is an interleukin that acts as both a pro-inflammatory cytokine and an anti-inflammatory myokine. IL-6's role as an anti-inflammatory myokine is mediated through its inhibitory effects on TNF-alpha and IL-1, and activation of IL-1ra and IL-10.


Tumor necrosis factor (TNF, tumor necrosis factor alpha, TNF-α, cachexin, or cachectin) is a cell signaling protein (cytokine) involved in systemic inflammation and is one of the cytokines that make up the acute phase reaction. The primary role of TNF is in the regulation of immune cells.


Cystatin C or Cystatin 3, a protein encoded by the CST3 gene, is mainly used as a biomarker of kidney function. It is a potent inhibitor of lysosomal proteinases (enzymes from a special subunit of the cell that break down proteins) and probably one of the most important extracellular inhibitors of cysteine proteases.


Serum Amyloid A protein-1 is a multifunctional apolipoprotein produced by hepatocytes in response to pro-inflammatory cytokines. It is secreted as a 12 kDa, 103 amino acid, nonglycosylated protein and circulates as part of the HDL complex. SAA1 can bind the surface of invading gram-negative bacteria, acting as an opsonin to aid clearance by macrophages.


Ly96 binds bacterial lipopolysaccharide. Cooperates with TLR4 in the innate immune response to bacterial lipopolysaccharide (LPS), and with TLR2 in the response to cell wall components from Gram-positive and Gram-negative bacteria. Enhances TLR4-dependent activation of NF-kappa-B. Cells expressing both LY96 and TLR4, but not TLR4 alone, respond to LPS.
