Introduction

Among the numerous genes in the genome, there are 20%-30% gene encodes the membrane protein. These membrane proteins are involved in almost every intracellular activities in life, including ligand-receptor binding, signal transduction, molecular transport, intercellular recognition and enzyme catalysis. Since the membrane proteins participate in such a large number of physiological activities, it's an important target for drug development and clinical research. As an important reagent in experimental research, the antibodies which against these membrane proteins are essential, however, there are many problems by using traditional methods to prepare these antibodies, including the low expression rate, hard extraction and purification, heterologous gene encoding, mis-folding, incomplete post-translational processing and conformational differences, etc.

Membrane proteins, actually, are a kind of proteins acting as ion channels, receptors and transporters, which enable the cells to transport environmental signals across the biological membranes. Membrane proteins can be classified into two categories, integral (intrinsic) and peripheral (extrinsic) membrane proteins—based on the nature of the membrane-protein interactions (Figure 1).

In principle, we use cell membrane, phospholipid membrane and liposome membrane to express the natural conformation of membrane protein in situ, this made the antibody we prepared has high fidelity and the ability to identify the characteristic structure of natural antigens. By now, the related experimental technology and process have been sent to the relevant department for patent examination, we overcame a series of difficulties of membrane protein antibody preparation, successfully produced membrane proteins covering a number of areas and a variety of functions.


Integral membrane proteins, have one or more segments that are embedded in the phospholipid bilayer, of which most span the entire phospholipid bilayer. These transmembrane proteins contain one or more hydrophobic membrane-spanning domains (α helices or multiple β strands), extending into the aqueous environment. Peripheral membrane proteins, do not interact with the hydrophobic core of the phospholipid bilayer. Instead, they are commonly bound to the plasma membrane by indirect interactions with integral membrane proteins or by direct interactions with lipid polar head groups. Peripheral proteins are located to the cytosolic face of the plasma membrane, playing a role in signal transduction. Other peripheral proteins, including certain proteins of the extracellular matrix, are localized to the outer (axoplasmic) surface of the plasma membrane.


It has been proven that membrane proteins perform a wide range of functions in cell growth, cell-cell communication (signaling transduction), differentiation, flow of information, metabolism, and migration. Defects in some membrane proteins can lead to diseases, such as cancer, therefore, membrane proteins represent nearly 50% of the targets of therapeutic research. It is very difficult to study membrane proteins due to their naturally low expression levels, although, the study of the structures of membrane proteins is a hot-spot issue all over the world because this will help to better understand the functions of these membrane proteins.
